Kintor Announces New Advanced Androgen Receptor Drug GT20029

China Trial Enrolls 180 Patients To Complete Enrollment: Update(8/22/23)

On August 22, 2023, Kintor Pharmaceutical announced the completion of patient enrollment in a phase 2 trial in China for GT20029. This means all of the patients needed to complete the study have been enrolled, and will undergo their dosing schedule over the next 12 weeks. The trial of GT20029, which is an androgen receptor degrader drug, is a multi-center, randomized, double-blind, placebo-controlled study designed to evaluate the efficacy and safety of GT20029 in treating male AGA, and to determine the recommended dosage for the phase 3 clinical trial.

Dr. Youzhi Tong, CEO of Kintor Pharmaceutical, said “We expect to release the top-line data of the Phase II clinical trial in the first quarter of 2024, and look forward to collecting more efficacy and safety data of GT20029 in this trial.

US GT20029 Trial Begins: Update (2/3/22)

Steady and exciting progress continues for Kintor Pharmaceutical. Early this morning, apress releasewas issued by Kintor announcing the dosing of the first subject in a phase 1 clinical trial for its innovative drug GT20029. See below for description of GT20029. The drug will be delivered in a topical gel form and is being trialed against androgenetic alopecia and acne vulgaris. Dr. Youzhi Tong, CEO of Kintor Pharma, shared these words in the release: “The start of this US clinical trial represents a significant milestone for Kintor, as it demonstrates the beginning of the international development of the company’s fourth innovative drug. In preclinical studies, we found that GT20029 can effectively block the androgen receptor pathway and its physiological function by degrading the AR protein. By limiting skin penetration, GT20029 inhibits local androgenic effects in peripheral skin tissues and so achieve good safety.

Bonus news: AClinicalTrials.govpage for a US phase 2 trial of Kintor’s other drug pyrilutamide has just been published. It appears that the trial is set to begin immediately. Three cohorts will test a 2.5 dose, a 5mg once and twice daily.

Kintor Pharmaceuticals of China announced through a press release today that the company is bringing yet another androgen receptor-based drug into clinical trials for the treatment of androgenic alopecia.

Androgen Receptor Degrader GT20029

Those who have been following along over the past year must be familiar withpyrilutamide, a topical androgen receptor-antagonist drug which is being trialed by Kintor in China and the USA for pattern hair loss. Pyrilutamide entered into a phase 2 trial in China at the end of 2020 and completed a phase 1 trial in USA last year for androgenic alopecia. The program is quite ambitious as Kintor has formally announced their plans to enter a phase 3 trial for pyrilutamide for AGA by the end of 2021, but it appears they’re not done yet either.

On February 2, 2021, Kintor announced that an investigational new drug application (IND) for the drug GT20029 has been approved by the NMPA of China (equivalent to the FDA) for the indications of androgenic alopecia and acne vulgaris. GT20029 is a new advanced form of an anti-androgen called a PROTAC which stands for proteolysis targeting chimera. It will be developed alongside pyrilutamide in the company’s pipeline.

A simpledefinitionof a PROTAC: “a heterobifunctional small molecule composed of two active domains and a linker capable of removing specific unwanted proteins.” Kintor states that GT20029 was developed through the company’s in-house PROTAC platform. Here are some explanatory statements by Kintor in their latestpress release:

“GT20029 is an AR degrader that degrades the AR protein. The mechanism of action of GT20029is to recruit the AR protein to the E3 ubiquitin ligase for degradation. It acts on the local tissues ofthe peripheral skin and local hair follicle sebaceous glands, which reduces the sensitivity of AR toandrogens without systemic exposure to the drug.

“As each PROTAC molecule can degrade multiple AR proteins, drugs based on PROTAC can achieve efficacy with a low dosage. In addition, as long as there are a small amount of PROTAC molecules in the cells, the efficacy of the drugs can be maintained, which can significantly reduce the dosing frequency as compared to other small molecule drugs.”

“According to animal experiments, the efficacy of GT20029 is superior to other small molecule AR inhibitors. In addition, GT20029 will not cause excessive drug accumulation and notable side effects. While achieving efficacy, GT20029 can effectively avoid systemic exposure to mitigate or avoid the side effects of the oral androgen signaling pathway inhibitors.”

Sounds good, yeah? This is the kind of drug evolution that most of us have been hoping for since finasteride became mainstream decades ago. It sounds like Kintor is getting closer to the ideal anti-androgen for hair regrowth and hair loss prevention (if all goes well). Kintor also states that they believe GT20029 is the first topical PROTAC drug which has entered the clinical trial stage anywhere in the world. An IND is expected to be filed soon in the USA for GT20029 as well.

    I’m digging Kintor’s efforts here and the fact that the company is China based is a plus for me, too. Chinese companies tend to be serious about their science and their business for sure. This new drug may be a bit down the line, but do not forget about pyrilutamide which is intended to reach phase 3 this year! Hats off to Kintor Pharmaceuticals (Pun fully intended).

      Finally!! a new compound in clinical trials! Hopefully it is more effective than finasteride and dutasteride, which although effective, do not eliminate the genetic burden of androgenic alopecia. Hopefully this compound is effective, especially for those of us who want to maintain our hair over the years, without having to go through more hair grafting operations. Unfortunately as an athlete I’m worried about the effects of nuking my DHT on my performance. An effective alternative to finasteride could come in the form of pyrilutamide or the drug which is highlighted in this very article, GT20029. Maybe 2024-2027 for the both of them, starting in China.

      Do you know if miniaturized hair follicles still have androgen receptors? Could this product reverse miniaturization if the receptors are “stuck on”? I’m wondering if increased sensitivity to androgen gets to the point where the receptor is just stuck on, which means a degrader could really help.

        Yes miniaturized follicles still have androgen receptors. AR’s are a natural part of a hair follicle, they are essentially a protein. GT20029 could potentially reverse miniaturization if it can remove androgens from the hair follicles.

    After first hand nasty experience with anti androgens, anything labeled AA scares me. They claim no systemic side effects but it’s just hard to believe. Who knows. The aa did work for me for decades and then the side effects came crashing down hard.

Fluridil has been great for me as a mild anti androgen. It has given me 0 sides and seems to maintain my hair. It’s binding affinity is also decent and better than breezula for Instance. Have you considered that?

      I totally agree Dale. I guess, if you are gonna be bald you have to just deal with that. I remember how my relative hoped in the beginning of 21st century that soon baldness would be cured. Two decades passed and I hear the same. It’s funny though that we are planning to go to Mars and on the Earth we have baldness problem. LOL.

          Hey PP, PRP is a ‘hit or miss’ type of treatment. It works decent for some and not for others. If money is no object to you it is definitely worth considering. I’ve heard recently from a scientist who works on PRP a lot that he believes it needs to be administered monthly for a bout 4 months in a row to be most effective, just a thought.

        GG—you are not discouraging others, but these comments are extremely unhelpful. I’ve mentioned this before to others, but Il keep going at it: judge updates on it’s merits and on the science. Don’t use fallacies like the argumentum ad antiquitatem to paint a dark picture of the future. Many things that seem simple are big issues in science. Hair loss is a very complex hormonal issue and the workings of the endocrine system isn’t well understood.

    @Ben: Sadly, I didn’t manage to participate but as it seems it was for potential investors only as Stemson is apparently going through the process of raising further funds for the upcoming pre-clinical studies.

    On another note, some of you may read it already but Riken has set up a Twitter account solely for the hair tooth organ regeneration segment on which they posted: “The novel study on the relation to clinical application of hair follicle regeneration will open at 10 AM (UK time) February 10, 2021. Coming soon.” Honestly speaking, the Twitter account does not seem to be the most reliable source but this could potentially be the first sign of life of the project in quite a while. Not getting my hopes up there either but after H2 2020 I guess any news is good news.

    I mean, the way they are building suspense here (“embargo time”) is making me curios.. I just hope they don’t announce they are going down the topical road with something that’s 5% more effective than Minox which Dr. Tsuji coincidentally found while doing research on cloning. Tomorrow could be a big day for the hairloss world, we’ll find out soon!

However, I’d also be happy with an improved Minox. It’s probably best, no matter how much they tease, to keep expectations low.

      Yeah Woofy, we’ve seen pictures of mice with hair years ago by other companies, really nothing new wit Tsuji.
      Because what happens in mice doesn’t always happen in humans.

      Tsuji still needs to provide us with physical proof of something in humans.

      I’m also expecting something more from Tsuji besides something slightly better than Minoxidil. I mean, there’s treatments in the pipeline that will be released sooner that are already claiming to have better results than Minoxidil, even TH16 claims to be better than Minoxidil and that’s a natural topical lol which is going to be released in Q1 this year!

      People would expect the highly regarded Tsuji, who people widely regard as one the the biggest names in the hair loss restoration game, to blow these smaller companies out of the water with their technology and treatments for hairloss.

      Someone regarded in such high esteem by the hairloss industry such as Tsuji should be releasing treatments FAR more effective than the others. He shouldn’t be planning on releasing small things the equivalent of some of the smallest companies in the pipeline.

      We’ll see soon enough if he really is worth the hype or if his just overrated.

    these drugs are cool but i dont see how they improve upon current solutions like accutane or minoxidil. they may have less side affects but their benefits are limited to the local tissues theyre applied to.

    acne vulgaris affects the entire surface area of your body, with the worst of it affecting your face and scalp where our sabacus glands are most abundant. youd hav to prescribe one topical for your body, another for your face, then another for your scalp just to meet the effectiveness that a systemic drug like accutane could give you with single pill a day.

    same for hair loss, topical min is a joke compared to taking it orally. which is why the first line of defence against hair loss is most countries is a daily combo of .5mg oral dutasteride with 10mg oral minoxidil. i hope this issue gets addressed before they hit markets.

    ^Agreed, maybe it would make more sense to combine the two articles? My response to this news is also the same: Kintor seems amazing, I am concerned about side effects. The press release statements seem to contain somewhat contradictor descriptives: “devoid of” and “mitigate” carry very different implications.

      It’s a good idea but the reason there were 2 articles in the first place was because Kintor has 2 drugs at the moment. I don’t favor the idea of a large number of updates in any article, so will keep it the same for now. Soon enough Kintor will have better news and I will publish a brand new article for it.

      Also, I’m interested to see how much GT20029 can mitigate side effects. They talk about only needing a small amount to be effective.

    I found something quite interesting.

    There’s a company called ARVINAS which is already developing a PROTAC Androgen Receptor Degrader which is already in Phase 2 Clinical Trials for their drug called ARV-110

    The only thing that I find highly strange is that it is only being trialed for Prostate Cancer by the company.

    We know that Anti-Androgen treatment is often used for Prostate Cancer treatment just like Finasteride is used alternatively.

    So my question is, do you think ARVINAS does not know about the Golden opportunity they have that this treatment can also be used as the solution for Androgenetic Alopecia ?!

    Do you think you could please reach out to ARVINAS and find out if they could start a trial for AGA? And if they know this PROTAC can also work for Male Patterned Balding, which from a business point of view for them, would mean killing 2 birds with one stone and obtaining double the amount of profits by treating 2 problems! (Hairloss and Prostate Cancer)

    It would be amazing because they might be able to actually release their PROTAC well before even Kintor Pharmaceuticals GT20029, since they have already finished their Phase 1 and soon their Phase 2 Clinical Trials!

    Its definitely worth a shot.

    This seems to say that Kintor’s new anti-androgen (GT20029) works topically and literally ZERO of the drug gets deeper than the local skin where it is applied so literally zero of GT20029 gets into the body. Is that correct? Could you please ask them about that because if literally zero of this drug gets deeper into the body then there is a zero percent chance of side effects.

      Russell, I think zero is unlikely, it’s more likely that a negligible amount gets into the body which could drastically reduce the potential of side effects and would be a big win for almost everyone.

      Russel, ZERO is impossible, it will never be. Even drinking too much water has side effects! Your body is programmed to work so that your hair falls out at a certain point. Fighting this is, by and large, an alteration of this. Now this all is about finding the right way to do it with the minimum possible sides.

      There will always be someone who can’t tolerate the treatment, someone who experiences little to no improvements and so on. That’s why is so important to widen the range of options to fight hair loss: if a treatment is not good for you, another may be. So far we have extremely limited cards to play…

    usually, what is the duration of phase 1 , 2 and 3 trails?

      Durations of phase 1 trials are usually within 4 months. Kintor could have already wrapped their phase 1 of GT20029 and is observing data and/or potentially doing follow up evaluations on subjects to ensure further safety.

      Progress is great but people who already lost all their hair and want them back are obviously not too interested in new maintenance drugs. I understand this sentiment. Fortunately, new maintenance drugs will come very soon, stronger growth stimulants probably too,
      and for many guys, this is fantastic news. Turning a Norwood 7 to a Norwood 0 is just possible by hair multiplication, but there are so many question marks. Will it cycle? Will it be safe? This is actually very important because the fda sees baldness as a cosmetic problem and will be very very strict on safety. Will it be dht-resistant? Can they control color? Can they control thickness? Curl? It will take a while to address all these questions.

    If Epibiotech / hairclone succeeds and their cells don’t loose inductivity and the treatment is deemed safe, cure could come within 2025. Even if the results are a 20% improvement on the intercytex results, multiple injection could in theory cure a NW5 easily.

      Dee—right Dee! And if we look at the last 44 years, quite some progress has been made. The hair transplants today are much much better than even 20 years ago. Also, finasteride and minox have been fantastic new medications. Now within the coming years, we might see the first FDA approved topical anti androgens as well.

      @BigL , none of the above. It competes with DHT for AR, and since its binding affinity is stronger than DHT (in theory) so it will bind with the AR hence DHT can’t bind with it.

I’ve heard some people saying that they’re tired of hearing the cure is almost here for the past 20 years. Is this still the case?       Others can feel free to share, but the simple answer is that the number of companies with potential treatments (i.e. shots on goal) is significantly higher than anytime over the last 40 years. Not to mention that the understanding of hair follicle biology has improved to some degree. There’s something like 46 companies listed in the Master article and I’d say around 15-20 are near term contenders. The application of science improves over time, it’s a validated statement.

      There are a lot of treatments in clinical trials, nothing like this happened 20 years ago, you can verify that yourself by googling “hair loss clinical trials” and filtering the reseach before 2005 or 2006. Just a couple of interviews to random people promising stuff. Quite different than actual companies researching actual medications, as it is now!

      People writing “guh, SaMe ThInG 40 yEaRs AgO”…yeah man, except for minoxidil, finasteride, dutasteride, hair transplants, palmetto, LLLT, PRP, etc…

    Obviously the admin knows much much more than me and has way better insights.
    I would say there was never that much research/attempts and the newest science is thrown at hair loss now. There will surely be effective topical anti-androgens out within 5 years complimenting the things we have now. Additionally, cell therapies are C
    currently being developed (like what hair clone is doing). I believe this might be offered within the next 5-10 years. Moreover I feel like hair transplant surgeons are really pushing the boundaries on what can be achieved with body hair, look up eugenix in India for extreme cases. Moreover, I feel like hair systems are losing their stigma amongst young people due to clever marketing by studios, at some point it won’t really have the toupee falling off joke from the 80s stigma anymore.
    The rest is speculation as nothing is in clinical trials today. Obviously hair cloning is the holy grail, but it is the hardest to figure out too.

      Ralph, I was referring to the difference in hair count increase from the Chinese trial to the US trial. I’d have to go back to recheck, but I think the numbers were like 22 in China to 10 in the US. No hyperbole in my statement, just stating that the expectations are a little different now. What’s your take?

        It’s a little disappointing that Epibiotech (Another S. Korean company) hasn’t mentioned anything about their clinical trials for EPI-001 considering they were meant to start in the 1st half this year. (I believe they are funded by the government too, unless I’m mistaken).
