US Male Phase 2 Pyrilutamide Trial Underway: Update (3/1/22)

Consistent progress is the M.O. of Kintor Pharma. On March 1, 2022 it wasannouncedthat the first patients in a US phase 2 trial for males have received dosing. The four cohorts in the trial will receive topical dosing of either 2.5 mg pyrilutamide twice per day, 5 mg pyrilutamide once per day, 5 mg pyrilutamide twice per day, or placebo over the course of 6 months. The estimated study completion date is January 2023.

Kintor Begins China Trial For Females: Update (11/12/21)

In its latest press release, Kintor Pharmaceutical has announced that the first patient has been dosed in a phase 2 double-blinded, placebo-controlled, all-female participated clinical trial for AGA. The study is set to include 160 female subjects who will receive doses of either pyrilutamide or control over a period of 6 months. Thisnewscomes as we await further detailed information from Kintor’s successful phase 2 trial of pyrilutamide in males with AGA which was projected to be released by the end of 2021.

FDA Approves US Phase 2 Pyrilutamide Trial: Update (7/11/21)

In a rare Sunday-issued press release, Kintor Pharmaceutical of China announced today that a phase 2 trial for their drug pyrilutamide will be taking place in the US shortly. The news stems from a recent approval Kintor received from the US FDA to begin the trial. The study, which comprises male subjects, will be randomized, double-blinded, and vehicle-controlled to observe a change in non-vellus hair count over a 6 month period.

Another announcement in today’spress release, equally as important, is pyrilutamide’s phase 2 trial for androgenetic alopecia in China has been completed and the data is expected to be read out in Q3 2021. Phase 2 trial data sets a strong precedence on how useful a drug may be for AGA. We should be finding out the important details by the end of October, let’s hope pyrilutamide impresses.

Perhaps most importantly, and an insight to the results achieved in pyrilutamide’s phase 2 China trial, Kintor also published a “Q2 2021 Business Update” earlierthis monthwhich stated that “a discussion on pyrilutamide’s phase 3 protocol for the treatment of androgenetic alopecia among principal investigators was held in Hangzhou, China.Remember, Kintor did sharewritten plansto start a phase 3 in 2021. A new androgen receptor-blocking drug may be on its way to the US and China for the treatment of hair loss. If successful in its current clinical trials, this drug could become an effective alternative to finasteride.

Suzhou Kintor Pharmaceuticals recently launched an initial public offering (IPO) in China which was met with a great response. The company is developing androgen receptor (AR) antagonist drug therapies for prostate cancer and pattern hair loss. Proxalutamide is Kintor’s AR antagonist drug indicated for prostate cancer which is currently in a US phase 2 trial and a phase 3 trial in China. Kintor plans to apply for marketing approval in China for proxalutamide before the end of 2020. Pyrilutamide is the company’s topical AR antagonist aimed at treating hair loss.

Here is a quote on the current progress of pyrilutamide from a recent mediaarticleon Kintor Pharmaceuticals:

The news about dual trials going on in the US and China for pyrilutamide is great, but the last sentence is even better. A phase 3 trial for pyrilutamide in the US is planned for next year (2021)? If this is true, it could be epic. One might now ask ‘if there is only currently a phase 1 trial for pyrilutamide in the US this year how could they get to phase 3 next year?’ Well, if the quote is true and not a miscommunication, the only logical explanation is the company plans to submit data from the China phase 2 to the US FDA and receive approval to go into a phase 3 trial. We will see how this develops in 2021. Another interesting point is the quote above alludes to the fact that pyrilutamide may be useful for both men and women. We have seen that Breezula, another AR antagonist drug, is likely to be useful for both genders.

Over The Next Year For Kintor

One question that immediately stands out in my mind is whether the company’s prostate cancer drug proxalutamide may also prevent hair loss or cause regrowth. It is an androgen receptor antagonist, after all. If its phase 3 trial goes well, proxalutamide is likely to be available on the market in China in mid-late 2021, so it will not take long to find out if it has hair benefits also. Another query I have is what the differences between proxalutamide and pyrilutamide are, this is not made clear through Kintor’s website.

Most importantly, we can expect the phase 2 trial results from Kintor’s hair specific drug, pyrilutamide, around the beginning of 2021. Let’s hope for good things.

  1. This looks like it could be a major breakthrough but I don’t understand how women could use it when any anti-androgen can cause birth defects. Has the company found a way to make this drug work only in skin without passing into the rest of the body? If they have then that would be a major accomplishment.

    • Well, we know that Cassiopea believed their AR receptor inhibitor drug was safe enough to launch a phase 2 trial for in women. In the future we will find out if Kintor is doing the same for pyrilutamide, beyond what was stated in the Chinese media article.

      • Also, Cassiopea’s acne (weaker) version is safe enough to use on children.

      Skin applying is really bad thing because it’s so inefficiently. The best if it would be as pills.

      • I think topicals can work equal and maybe even better than oral in the right circumstances (setipiprant?). A lot of people sometimes prefer topicals. The proxalutamide drug is oral so maybe there will be something with that.

  2. I’ve been searching for details about this drug. My issue with it is that the company wants to market it to woman as well as men. But anti-androgens can be very harmful to fetuses and cause serious defects. That’s why Merck never pushed finasteride to treat female pattern hair loss. Women are not even supposed to touch finasteride pills. Men who are using finasteride are not supposed to have women who plan to bear children. So I don’t understand how women could possibly use Kintor’s anti-androgenic drug for hair loss. If even the tiniest amount gets into a woman’s body it could be devastating to a fetus.

    • How are you certain that the company wants to market this toward females? So far their trials are only in men. By the way, I’ve heard of reputable hair surgeons prescribing finasteride to women, most likely when they are past the child birthing phase.

      To your question, I am not sure. I am mostly certain the pyrilutamide is a separate molecule from proxalutamide, basically because if they were the same drug I think the company would be more inclined to just call pyrilutamide “topical proxalutamide.” Anyways, cool AR stuff coming out of China. I’m hoping for a winner here.

  4. There is another androgen 5α-Reductase and it is used to treat prostate illness. I had it about a year and it is absolutely inefficient. I had a bit new hair growing absolutely unnoticeable if you don’t know about it. And I had new hair are grown on the sides of my lips and it’s really annoying and constant effect.

  5. The potential dangers of topical finasteride, which alters hormonal levels, do not carry over to topical receptor blockers like Breezula and now Pyrilutamide, which is why they are expected to be safer for everyone.

    The reference is on p. 32: “Clascoterone Solution could be priced like other cash pay lifestyle drugs ie $100-200 per month.” I am not sure what a “cash pay lifestyle drug” is, but surely they’re not thinking about Rogaine? I only use half the recommended dose, but still I can get the foam version for only $15 a month, which is nowhere near $100.

How much do people pay for Propecia prescriptions, anyone here care to share? And does your insurance company pay for any of the cost? Because there’s almost no way Cassiopea can overprice their drug too much and expect to be competitive if their drug is not markedly superior to finasteride at this point.

      • Yoda can testify to you lads that in the 80’s when 2% Rogaine was a prescription item it was a heck of a lot more expensive than it is now. My brain is telling me that it was $50 for 60 ml. It may have been more but definitely not less.

    • Has anyone tried dutasteride mesotherapy? Dr. Carlos Wesley in NYC is offering it and claims it need only be done once every 3 months and the method essentially eliminated systemic absorption limiting the drug to the local area of the scalp.

  13. I saw the below paragraph at a different site. The paragraph concludes that the Kintor anti-androgen doesn’t get into the body when applied topically.

    “A topical AR antagonist, pyrilutamide is designed to address the shortcomings of minoxidil and finasteride. It is being developed to locally block androgen-mediated signaling instead of reducing androgen level systematically. Its metabolite has substantially reduced AR agonist activity in vivo, thereby limiting its side effects. In comparison, minoxidil may trigger an allergic reaction to propylene glycol as well as orthostatic hypotension, while finasteride may lead to adverse sexual side effects.”

    Can someone please tell me what statement inside of this above paragraph proves that Kiintor’s anti-androgen does not get deeper into the body when applied topically. It looks like they’re saying that since “Its metabolite has substantially reduced AR agonist activity in vivo, thereby limiting its side effects.” this somehow means that it doesn’t get deeper into the body when applied topically. How does the statement that its metabolite “substantially reduced AR agonist activity in vivo” mean that it doesn’t get into the body when applied topically?

    Can someone smarter than me please help me understand this?

    • Russsell,

      They are saying the drug has antagonist abilities when applied topically, so it blocks the androgen activity. But once it starts to degrade, the metabolite (or the new degraded compound) has low agonist (or mimicking) androgen abilities. So from the moment you apply the drug to when it would reach other parts of the body, the drug has already been broken down into a smaller compound that no longer has antagonist effects, but in fact acts as an agonist now.

      To be quite honest, It would make more sense if they meant the metabolite has reduced “antagonist” effects but perhaps the low agonist levels also helps to maintain appropriate androgen levels.
      This part is a bit odd to me. I’m a biochemist but that’s not really my area of research, so perhaps someone else can chime in.

      • My guess in response to Sierra is that low agonist ability is saying that when the metabolites are produced they do not seem to have affinity for the active site of the A receptor around the body.

Before corona there were months December 2019 – March 2020 to start the trial and they didn’t start… can’t explain this and I am wondering that Jon Knight didn’t give you a reasonable statement on this because in 2019 Jon Knight was very polite and open and now he is calm and don’t talk to you? In summary, J. Hewitt is still seeking a CRO partner to run the trial, if and when they find one most likely there will be a big update about it, wish them well.

  18. 你好管理员,I had this query I had to ask you. I am 17 right now and I feel like my hairline is receding near my temples(it is still very very less noticeable). I just want to treat it early than after loosing most of it. And I have a family history of baldness on my father’s side. Should I wait for the future treatments or should I start on minodixil like how my dermatologist said? Because he just prescribed it without any other advice and I don’t think my is that bad that I have to start it.

    P.S- Sorry for asking this. I just wanted some advice from someone who knows about these things and you seem to know about this

    • Hi Shaan, I would suggest you follow the advice of your dermatologist. Minoxidil seems like a good bet for you and could help delay the progression of your hair loss until future treatments come out.

In my opinion it also augments the other news we get about new companies/startups, it’s good to have stuff in the near term and further development timelines.

  23. Any idea how those kind of drugs, when sucessfully developed, may affect the raise of hair cloning with all its possible approaches?
    It will save the day for a lot people (or even majority – with balding persons yet to come).
    On the other hand, even if one could benefit from it, it’s still a matter of taking medication till the rest of your life, right? The argument for cloning would still remain in the game.

  24. In my layman perspective this is the most significant development/company in AGA in awhile. I expect this drug to succeed clinical trials, I find it more dubious that this is a drug completely free of systemic side effects.

    • I also don’t believe this topical from Kintor will have no side effects. They said the same thing about Finasteride that it would have no side effects until millions of people over the years found out the hard way that there are side effects.

      • Yoda was around for the release of finasteride/propecia in the 90’s…they never stated that it would be free of side effects, that is just “fake news” as some douche bag used to say. When I started on fin the doc was very upfront with the potential side effects as was the marketing of propecia.

        • Yes you might have been fortunate but many doctors, dermatologists and specialists often don’t tell patients the side effects as they feel it might cause them to overthink. I personally was prescribed Finasteride from a Dermatologist who didn’t tell me the side effects. I’m sure many people also have had such an experience. It’s common also on the internet where people tend to say that Finasteride side effects aren’t real or that its super rare to the point people like to make others it’s non-existent. My statement previously is based on the above.

Not sure how one would quantify “many”, there is much supporting scientific data to the contrary. Nevertheless, everyone is entitled to their own opinion based on their individual experience.

  25. I remember when they came out with propecia in the late 90s. I remember watching the commercial and they started erectile dysfunction and libido loss as the side effects for less than 3%. Then Jay Leno made a few jokes about propecia on the Tonight Show lol. He was like “you’ll get your hair back and women but you’ll lose your erections…can’t get a break” it was something along those lines Haha. Hey Yoda do you remember those commercials and Jay Leno making those jokes in his monologue?

    • Hey there Mr. Jones! I don’t remember Leno specifically doing that bit. However, I know those same sort of jokes generally made their way into popular culture. It was a fairly well known fact that Propecia had the (low) potential to cause sexual side effects in some based on clinical trials. As you related, this was also made clear in the marketing as well as on the packaging. Odd that some report that their docs didn’t educate them, sometimes 5 minute medicine can limit the information one gets. On the other hand, I’ve never been able to leave a legitimate pharmacy without the pharmacist or pharmacy tech educating me on the drug and also with a handful of information they provide on same.

  26. I was also prescribed finasteride by a dermatologist who was quite dismissive about potential side effects, which I had the misfortune of experiencing very quickly (horrible hot flashes!). I suppose we are now off topic. I hope that pyrilutamide will be a potential replacement for those who cannot tolerate fin.

  28. It takes critical thinking to follow progress Birdin. Obviously, situations will seem similar because every drug therapy has to go the same route of phase 1, 2, and 3. Some drugs we have seen don’t make it through, there will be different outcomes for other drugs. Find ways to bolster your own reasoning and faith in a positive outcome, it’s understood that some good things in life require patience. In regards to your last 2 sentences, projecting your momentary despair doesn’t usually help the process in the long run ? Detachment and other support systems help, e.g. a Spiritual practice.

    • The admin was being kind in his words to you. If you are so hesitant of a future treatment/cure you should remove yourself from this website. It’s not easy coming up with a solution as hair regeneration is a very complex problem. Imagine if everyone was so negative about a cure or invention like you because of a few decades of no true solution. In that case we’d have almost no cures or inventions that we have today.

      Some cures for diseases and some inventions were made earlier because it wasn’t as complex, but the good thing is that technology and medical inventions are growing. For example, we have Gene Editing which has recently been used in humans and proven to work. That was not possible before. Where we will be in the next 5 to 10 years time once we improve thar technology will be amazing for not just humans, but animals, and plants.

      Maybe the hair cures are not in your timeline, but the world doesn’t revolve around your timeline. I strongly believe we are closer than ever to a treatment and cure and the fact that Kintor is flying through the clinical trial process in unprecedented time is proof of that.

  31. Late on starting US Phase II trial (for men) – 4 months ago, they said it would start ‘soon’.

    Late on Phase II results (China). And they said they would be starting Phase III by the end of this year (in China). We’ll see.

    C’mon, Kintor – don’t turn into all the others that can’t keep their own deadlines. Up till now, they had been the one reliable one.

    • I’m not concerned about late on starting a trial in a different continent. However, waiting on the phase 2 results has a slight odd feel to it.

Imagine there is a treatment that work and is marketed but you have to wait 2 or 3 more years until the european agencies evaluate it ?     • being a nw7… dont expect much mate. antagonizing the androgen receptor will not create new hair follicles. you may grow back 1 norwood as a nw7 depending on how long ago it was you went nw6–>nw7. Reglardless, your hopes should only life with hair cloning because that is the true path to restoring a healthy head of hair for you

          I like how hair systems look but I don’t find the whole glue aspect attractive. It just doesn’t feel like it’s my hair that I can be proud it. I’m sure they have a time and place

          • 17 year old–Understandable. There are some downsides, but many of them are psychological barriers. Even pride is something worth examining; is pride really what you should be after? Perhaps being happy with how you look is more important? For example, I had a hair transplant and now I am much happier with how I look. Perhaps a hair system can do the same for you?
            Also, there are many positive sides to it! No expensive products, no pills etc.
            Anyway, wish you all the best

        • Thats true Josh. I know many guys dont go after transplants or hair systems because of their own pride. I find it a bit more challenging because if I go to class or home with a hair system .. my friends and family are gonna be asking a whole lot of questions. At that point, it just gets demoralizing.

          Hair transplants invoke more enthusiasm from me. I hope i can maintain my thick donor region. Its been pretty stable since i started thinning at the top.

        • Mike, I don’t have any experience with it myself. I know there has been one published study on it that Dr. Sergio Vano was involved in. I believe it can limit the systemic absorption but it cannot eliminate it.

        • Thanks for the interesting share, Sonic. To be honest, I would expect that companies who already have a therapy developed pretty far down the line are not going to make any important tweaks to it, but you never know. New therapies can always be developed down the line.

I have never experienced any real sides from minoxidil or finasteride, although I do take testosterone hormone therapy, so that may counteract some of the sides, perhaps the T is counterbalancing the Fin. But minoxidil and fin seem to have stopped my hair loss at a little over a Norwood 2, not quite a Norwood 3, but close, like a 2+ but still enough hair to avoid shaving the head, and not enough to have the strong hair line we all miss from our 20s and 30s..

    What is taking so long?

    I don’t experience finasteride sides, but many people on this forum do. And 50% of men experience hair loss by 50, 60% by 60. I imagine any man with thinning hair, like myself, would gladly take something with zero sides that not only stopped hair loss but reactivated growth.

    The target market is simply enormous. I don’t understand why the drug pipeline for a cure is moving this slowly.

    Anyone have thoughts on this?

    • Dee, this is just my personal opinion – from the outside looking in, it seems to be an issue of funding. Often times we see these biotech companies initiate their early level research in partnerships with universities. Seemingly, since the seriousness of hair loss has never been taken as largely as it is today the funding for this type of research has never been there. With the social media centred society we have today – I believe the funding has been improving as of late and we can see that will all of the new startups and research going on worldwide (especially in Korea where beauty is important for status.

    • Dee, I’d also mention that several attempts have been made over the last 6 years and some drugs just did not meet their endpoints in trials; setipiprant, bimatoprost, FOL-005, SM04554, and HST-001 all received millions of dollars in development funding but did not make the cut.

    Just curious about clinical trails for different countries. If a clinical trial is taking place in China, can those results be used for clinical trials in Canada? Or does Canada or U.S. need to conduct their own clinical trials before allowing commercial use of a new hair loss drug?

    • Gnarly, I don’t know specifically all the facts for this question. I know that some trials from other countries can be used towards a clinical trial path in the US, but approval in another country will not lead to direct approval in the US. For example, I remember RiverTown was going to do a phase 2 in Australia and use that to get into a phase 3 in the US. The phase 3 was definitely going to have to take place in the US.

    • Jose, I understand that perspective a lot and it makes sense. I think many people hope that pyrilutamide will be in the realm of finasteride. Being an equivalent is a tall order.

      Who’s this “doctor” and how can he know that if this drug is still under developement and the company that is researching it is being super secretive?
      Btw, even the mere maintenance without sides would be a great result!

    In your opinion, what is the best case scenario for Epibiotech’s cell therapy? I feel like people should be more hyped for this treatment. Replicel’s cell therapy was disappointing to many, but was still able to halt further progression of hair loss for 8 months with just one shot, if I’m remembering correctly. From what I understand, Epibiotech uses a similar but more advanced method of culturing the cells. I would think even a small improvement over what replicel did would be a game changer, right? Something that halts progression and maybe even regrows a little hair without altering hormones? I’m at least excited that they’re moving towards trials. Even if it doesn’t come to market for a few years I’m excited to see what they can do.

    • Mr. Burns, I think that Epibiotech’s cell therapy is one of the most interesting therapies in the pipeline at the moment. I don’t mind if people don’t want to be hyped because nothing is promised and some people handle the letdowns better than others. But, I do think the potential of EPI-001 is above average. I think a starting point goal for any new therapy is finasteride type efficacy without the potential side effects, if it can exceed that, then also great.

    I have good expectations on this drug and this company but this delay and this total absence of any data whatsoever is starting to be alarming…

    • Hollywood, it was some time ago that Kintor switched their strategy and began telling people via email response that they are waiting to release the China phase 2 results at a conference in mid 2022. The exact quote is floating around in the comments of one of these articles, but that is the gist of it. I don’t find that to be an overwhelmingly good sign, but it’s not necessarily a negative sign either. We will see the data results in mid 2022 and hopefully some photos included.

      • I thought they used it on the back at the trails? I dont think there ll be photos because of this. I hope so much kintor can stop hairloss from going on. They are legit my only hope…. If they fail i feel like thats it for the next 15 years.

        • Used it on the back? No I believe it was used on the scalp. They had to have taken photos, that is the way to determine efficacy.

          • Ah ok idk why i thought on the back and arms…. I thought they said they used totally healthy people without hairloss and tried it on areas which are easy to see by the docs. Thats why they know skin redness is the only sideeffekt?

            But hopefully u r right. I wanna see pics as well
